The discovery and engineering of novel fluorescent proteins (FPs) from diverse organisms is yielding fluorophores with exceptional characteristics for live‐cell imaging. In particular, the development of FPs for fluorescence (or Förster) resonance energy transfer (FRET) microscopy is providing important tools for monitoring dynamic protein interactions inside living cells. The increased interest in FRET microscopy has driven the development of many different methods to measure FRET. However, the interpretation of FRET measurements is complicated by several factors including the high fluorescence background, (...) the potential for photoconversion artifacts and the relatively low dynamic range afforded by this technique. Here, we describe the advantages and disadvantages of four methods commonly used in FRET microscopy. We then discuss the selection of FPs for the different FRET methods, identifying the most useful FP candidates for FRET microscopy. The recent success in expanding the FP color palette offers the opportunity to explore new FRET pairs. (shrink)

There is, however, no widely agreed definition of community engagement or of approaches adopted. Justifications given for its use also vary. Community engagement is, for example, variously seen to be of value in: the development of more effective and appropriate consent processes; improved understanding of the aims and forms of research; higher recruitment rates; the identification of important ethical (...) issues; the building of better relationships between the community and researchers; the obtaining of community permission to approach potential research participants; and, the provision of better health care. Despite these diverse and potentially competing claims made for the importance of community engagement, there is very little published evidence on effective models of engagement or their evaluation.MethodsIn this paper, drawing upon interviews with the members of a Community Advisory Board on the Thai-Myanmar border, we describe and critically reflect upon an approach to community engagement which was developed in the context of international collaborative research in the border region.Results and conclusionsDrawing on our analysis, we identify a number of considerations relevant to the development of an approach to evaluating community engagement in this complex research setting. The paper also identifies a range of important ways in which the Community Advisory Board is in practice understood by its members to have morally significant roles and responsibilities beyond those usually associated with the successful and appropriate conduct of research. In many applications of physics, boundary conditions have an essential role. The purpose of this paper is to examine from both a historical and philosophical perspective one such boundary condition, namely, the no-slip condition of fluid dynamics. The historical perspective is based on the works of George Stokes and serves as the foundation for the philosophical perspective. It is seen that historically the acceptance of the no-slip condition was problematic. Philosophically, the no-slip condition is interesting since the use of the (...) no-slip condition illustrates nicely the use of scientific models. But more importantly, both the use and justification of the no-slip condition illustrate clearly how theories can holistically approach the world through model construction. Further, since much of the debate over scientific realism occurs in the realm of models, a case is made that an understanding of the role of the no-slip condition has something to offer to this debate. In March 1928, John Dewey responded to a request from Marie Meloney, editor of the New York Herald-Tribune Sunday Magazine, and offered his recommendations on recently published texts on education. Dewey wrote, "I think the best educational books of recent publication are Bode, Modern Educational Theories . . . Since January 2016, the Mahidol Oxford Tropical Medicine Research Unit has trialled a data-sharing policy where requests to access research datasets are processed through a Data Access Committee. In this paper, we share our experiences establishing data management systems and data-sharing infrastructure including a data-sharing policy, data access committee and related procedures. We identified a number of practical and ethical challenges including requests for datasets collected without specific or broad consent to data sharing and requests from pharmaceutical companies for data (...) to support drug registration applications. We also encountered significant resource constraints which required the development of appropriate human resources and infrastructure. We suggest a research agenda to promote responsible and equitable data sharing while safeguarding the rights and interests of research participants and researchers. (shrink)

Back in the bad old days, it was easy enough to spot non-cognitivists. They pressed radical doctrines with considerable bravado. Intoxicated by the apparent implications of logical positivism, early noncognitivsts would say things like, "in saying that a certain type of action is right or wrong, I am not making any factual statement..." (Ayer 1936: 107) Like most rebellious youths, non-cognitivism eventually grew up. Later non-cognitivists developed the position into a more subtle doctrine, no longer committed to the revisionary doctrines (...) associated with its forefathers. For example, Simon Blackburn has undertaken the "quasi-realist" project of showing how a non-cognitivist can "earn" the right to the seemingly realist discourse on a less metaphysically controversial and semantically implausible basis by giving a non-cognitivist analysis of realist-sounding semantics and pragmatics (Blackburn 1993). Formalised knowledge systems, including universities and research institutes, are important for contemporary societies. They are, however, also arguably failing humanity when their impact is measured against the level of progress being made in stimulating the societal changes needed to address challenges like climate change. In this research we used a novel futures-oriented and participatory approach that asked what future envisioned knowledge systems might need to look like and how we might get there. Findings suggest that envisioned future systems will need (...) to be much more collaborative, open, diverse, egalitarian, and able to work with values and systemic issues. They will also need to go beyond producing knowledge about our world to generating wisdom about how to act within it. To get to envisioned systems we will need to rapidly scale methodological innovations, connect innovators, and creatively accelerate learning about working with intractable challenges. We will also need to create new funding schemes, a global knowledge commons, and challenge deeply held assumptions. To genuinely be a creative force in supporting longevity of human and non-human life on our planet, the shift in knowledge systems will probably need to be at the scale of the enlightenment and speed of the scientific and technological revolution accompanying the second World War. This will require bold and strategic action from governments, scientists, civic society and sustained transformational intent. (shrink)

Research funders, regulatory agencies, and journals are increasingly expecting that individual-level data from health research will be shared. Broad consent to such sharing is considered appropriate, feasible and acceptable in low- and middle-income settings, but to date limited empirical research has been conducted to inform the design of such processes. We examined stakeholder perspectives about how best to seek broad consent to sharing data from the Mahidol Oxford Tropical Medicine Research Unit, which implemented a data sharing policy and broad consent (...) to data sharing in January 2016. Between February and August 2017 qualitative data were collected at two sites, Bangkok and the Thai-Myanmar border town of Mae Sot. We conducted eighteen semi-structured interviews. We also conducted four focus group discussions with a total of nineteen people. Descriptive and thematic coding informed analysis of aspects of data sharing that are considered most important to inform participants about, and the best ways to explain complex and abstract topics relating to data sharing. The findings demonstrated that clinical trial participants prioritise information about the potential benefits and harms of data sharing. Stakeholders made multiple suggestions for clarifying information provided about data sharing on such topics. There was significant variation amongst stakeholders’ perspectives about how much information should be provided about data sharing, and it was clear that effective information provision should be responsive to the study, the study population, the individual research participant and the research context. Effectively communicating about data sharing with research participants is challenging in practice, highlighting the importance of robust and effective data sharing governance in this context. Broad consent should incorporate effective and efficient explanations of data sharing to promote informed decision-making, without impeding research participants’ understandings of key aspects of the research from which data will be shared. Further work is required to refine both the development of core information about data sharing to be provided to all research participants, and appropriate solutions for context specific-challenges arising when explaining data sharing. Since its discovery in 1912, X-ray crystallography has become a most useful tool in physics, chemistry, material science, mineralogy, metallurgy, and even in the biological sciences. In 1914, Max von Laue was awarded the Nobel Prize “for the discovery of X-ray diffraction by crystals,” followed by the 1915 Nobel Prize to William Henry Bragg and William Lawrence Bragg “for their services in analysis of crystal structure by means of X-rays.” And these early Nobel prizes marked only the beginning of X-ray (...) crystallography as one of the most fruitful methods in the history of modern science. This article highlights that in our rush to call for the democratization of science and expertise we must not forget to speak to what type of democratization we are calling for. In short, not all participatory forms are the same. In developing this argument, I examine one such form that has yet to receive much attention from science and technology studies scholars: the agricultural field day. In examining the field day, we find that its orientation—that is, toward either the conventional (...) or sustainable model of agriculture—shapes significantly its democratic approach to knowledge coproduction. To help inform how we understand conventional and sustainable field days, I turned to two conceptual frameworks of participation: namely, that of Wynne and Funtowicz and Ravetz. In doing this, we find that conventional field days mirror closely that form of public participation detailed by Funtowicz and Ravetz, while sustainable field days reflect that conceptualized by Wynne. Recent and rapid technological developments on many fronts have created in our society some extremely difficult moral predicaments. Previous generations have not had to face the dilemmas posed by, for example, the availability of safe abortions, sperm banks and prostoglandins. They have not had to come to terms with an unchecked exploitation of natural resources heralding imminent ecological crisis, or, worst of all, with the recognition that only in this current generation have people the capacity to destroy themselves and their (...) environment. This book seeks to show how, and why, Seventh-day Adventism has addressed these moral issues, and that the ethical questions arising from these issues are especially relevant to the Adventist church and its development. Dr Pearson looks specifically at the moral decisions Adventists have made in the area of human sexuality, on such issues as contraception, abortion, the role and status of women, divorce and homosexuality, from the beginnings of the movement to 1985. He seeks to put such decision-making in perspective by providing the general social context in which it took place, and shows how Ellen White (whose charismatic leadership held the movement together in its first fifty years) has been a major source of moral authority in the Adventist church - her writings continuing to exercise authority in a contemporary society of turmoil and change. This important book, which conveys something of the general ethos of Adventism, is the first to investigate the ethics of the movement, ans so fill a notable gap in the literature. (shrink)

Genetic engineering is often looked upon with disfavour on the grounds that it involves "tampering with nature". Most philosophers do not take this notion seriously. However, some do. Those who do tend to understand nature in an Aristotelian sense, as the essence or form which is the final end or telos for the sake of which individual organisms live, and which also explains why they are as they are. But is this really a tenable idea? In order to secure its (...) usage in present day ethics, I will first analyze the contexts in which it is applied today, then discuss the notion of telos as it was employed by Aristotle himself, and finally debate its merits and defend it, as far as possible, against common objections. Those who find their work meaningful often need to be more committed. Over-commitment, in turn, frequently results in stress, personal conflicts, and burnout. Such over-commitment, in other words, leads to employees needing to take more care of themselves. This paper considers the prospects for meaningful self-care in the context of working time reduction. For this, we consider the case of the four-day workweek, asking employees of such organizations to explain how they make meaning out of their newly found time off. (...) Conceptually, we rely upon the work of Michel Foucault, particularly his analysis of the care of the self. On its basis, we coded five self-care practices: (1) rest and recuperation, (2) professional and personal development, (3) domestic work, (4) balancing work, and (5) additional work. We conclude by highlighting the theoretical and practical implications of work reduction for the analytical, ethical, and practical pursuit of meaningful work. (shrink)

It is controversial how Levinas understands the interrelation between the ethical and the political. In this article, I propose an interpretation of that relationship and then provide a reading of the film Two Days, One Night, written and directed by Luc and Jean-Pierre Dardenne, in order to enrich that proposal. Bringing into focus how a film expresses developments in philosophical thought is a delicate and subtle matter. I hope to show how a careful reading of the film and in particular (...) the interpersonal, moral, and social dynamics that it exhibits can help us to see how, in a concrete setting, a rather abstract philosophical understanding of our engagements with others has social and even political implications.
